Single cell/nuclei RNA-seq

Single-cell RNA sequencing (scRNA-seq) enables the measurement of gene expression at the level of individual cells, revealing cellular heterogeneity that is often masked in bulk RNA sequencing experiments. This approach allows researchers to generate high-resolution maps of cellular diversity and gene regulation across complex tissues and biological systems. 

The CGI offers both scRNA-seq and single nuclei RNA-seq (snRNA-seq) library prep and sequencing services on both Illumina and PacBio instruments..   

Sc/SnRNA-seq is performed in the CGI using Illumina Single Cell library preparation kits (formerly Fluent PIP-seq technology). User supplied single cell suspensions or nuclei isolates can be processed for scRNA-seq and sequencing using the Illumina workflow. We also carry a Singulator 200+ for nuclei and whole cell isolation from tissues, cell suspensions, or FFPE samples. 

Several configurations are available for sc/snRNA-seq based on your sample size and complexity.

The PacBio Kinnex single-cell RNA kit utilizes MAS-Seq, an array concatenation method for increasing throughput by joining cDNA molecules from single cell RNA-seq library preparation workflows like 10x Genomics, into longer concatenated fragments bioinformatically. PacBio HiFi reads generated from single cell cDNA library preparation products can then be bioinformatically parsed to retrieve the original cell-specific molecules all maintaining synthetic long reads. 

The Kinnex kit is compatible with cDNA generated from most single cell platforms including Illumina Single Cell library preparation kits (formerly Fluent PIP-seq technology; offered in the CGI, or user prepared), and other single cell cDNAs prepared by users with Parse WT and WT mini kits, and 10x Chromium Next GEM Universal 3’ (v3.1), Next GEM Universal 5’ (v2), GEM-X Universal 3’ (v4), and GEM-X Universal 5’ (v3). 

Recommended input is 15–75 ng of cDNA.
